# MT-GNN: Predicting Brain Morphometry with Mesh Evolution in Continuous Time
## Overview
MT-GNN (Mesh Tensor Graph Neural Network) is a novel approach for predicting how subcortical brain structures' shapes will evolve over time from multiple prior scans. Instead of traditional methods that either use high-dimensional embeddings or directly regress vertex deformations, MT-GNN predicts the surface's intrinsic geometry in continuous time by forecasting the per-vertex first fundamental form (metric tensor).
## Key Innovation
- **Intrinsic Geometry Prediction**: Predicts metric tensors instead of direct vertex positions
- **Continuous Time Modeling**: Handles arbitrary prediction horizons with Fourier encoding of lead time
- **Differentiable Reconstruction**: Uses As-Rigid-As-Possible (ARAP) solver to decode metric into valid surfaces
- **End-to-End Training**: Trains on rigid-aligned vertex error through the reconstruction pipeline
## Performance Results
- **Superior Prediction Accuracy**: Outperforms all evaluated methods at every prediction horizon
- **Consistent Improvement**: Beats temporal mean baseline on all 14/14 subcortical structures tested
- **Advantage Over Competitors**:
- Ahead of geodesic shape regression (DCM)
- Better than mesh transformer (TransforMesh)
- **Horizon Scaling**: Performance advantage widens as prediction horizon increases
## Technical Components
### Graph-Based Metric Tensor Embeddings
- Single per-structure graph network architecture
- Predicts future per-vertex first fundamental form (metric tensor)
- Conditioned on Fourier encoding of lead time
- Handles arbitrary causal multiple-visit history
### Differentiable Surface Reconstruction
- As-Rigid-As-Possible (ARAP) solver for metric-to-surface conversion
- Ensures decoded predictions remain valid surfaces
- End-to-end differentiable training pipeline
- Rigid-aligned vertex error as training objective
## Applications
- **Clinical Prognosis**: Predicting disease progression from limited scan history
- **Clinical Trial Enrichment**: Identifying patients likely to show morphometric changes
- **Longitudinal Analysis**: Understanding brain structure evolution patterns
- **Personalized Medicine**: Patient-specific morphometry trajectory prediction
## Implementation Guidelines
### Data Requirements
- Multiple time-point scans of subcortical structures
- Surface mesh representations with consistent topology
- Rigid alignment across time points for training
### Architecture Design
1. **Graph Network**: Implement per-structure GNN for metric tensor prediction
2. **Fourier Conditioning**: Encode lead time using Fourier features
3. **ARAP Solver**: Integrate differentiable surface reconstruction module
4. **Loss Function**: Use rigid-aligned vertex error for end-to-end training
### Training Strategy
- Train on ADNI dataset or similar longitudinal neuroimaging data
- Validate across multiple subcortical structures (14 structures in original study)
- Test performance at various prediction horizons
- Compare against temporal mean, DCM, and TransforMesh baselines
## Evaluation Metrics
- **Mean Vertex Error**: Primary metric vs. ground truth surfaces
- **Temporal Mean Comparison**: Baseline performance comparison
- **Structure-wise Analysis**: Performance across different subcortical structures
- **Horizon Analysis**: Performance scaling with prediction time
